The founding of Porto Murtinho is linked to the Companhia Matte Larangeira: in 1892, a river port was installed at the Três Barras Farm for the shipment of the yerba mate produced in the region, named in honor of Joaquim Murtinho. The settlement developed with the yerba mate cycle, was elevated to a municipality in 1911, dismembered from Corumbá, and was part of the extinct Federal Territory of Ponta Porã between 1943 and 1946.
The economy of the municipality is based on agriculture and livestock farming — with one of the largest cattle herds in the state —, fishing and tourism. In 2023, the gross domestic product (GDP) of the municipality was R$571.5 million. Porto Murtinho is the Brazilian starting point of the Bioceanic Route (Capricorn Bioceanic Corridor), a road route that will connect the Atlantic Ocean to the Pacific Ocean through Paraguay, Argentina and Chile, whose international bridge over the Paraguay River was in the final stage of construction in 2026.
